There are rest stops/service plazas along the highway, and you can buy them there. If you are going into switzerland, and you forget to buy one, you can get one at the border after they check your passport. The guard just asks you to pull over to the side and there is an agent with a money belt and coin dispenser whose only job is the toll sticker.

Well, here is how not to do it: as you pass the last exit before entering Austria, miss the ramp, stop on the Autobahn, and then back down the entrance ramp.
Otherwise, you can buy it at almost any gas station near the border.

I found when going into Austria many of the large gas stations right off the autobahn had them. If I remember correctly, there were also signs on the autobahn that showed the gas station's name and indicated they sold the vignettes. I picked mine up at a Shell station just accross the German boarder near Innsbruk.

I have not experienced firsthand, but have read here and in the travel books that they can be purchased at most gas stations near the borders. Also, have heard strong advice to purchase them, the ticket for not having one is quite expensive.

FWIW, the vignettes are only required when driving on the autobahn's. For B-road driving, no vignette is required in either Austria or Switzerland.
The rest-stations that sell the vignettes usually will be identified by an icon on the sign announcing the rest station, and start about 20 miles from the respective borders.
